Not a bad seat in the house but if you want the best seats possible in the 300 level sections, get the box seats and save yourself from climbing all of those stairs. Plus, there are only five or six rows of seats as opposed to 20+.
Sitting behind home plate will allow you to see both the Detroit skyline and the giant scoreboard without adjusting your position in your seat to see either. I thought the two Tigers on top of the scoreboard were a nice touch as are the fountains in the outfield.
But if you're coming to Comerica for the first time, don't just walk to your seats. Do a lap around the bottom level and soak in the sights and sounds, Comerica has a ferris wheel and carousel for the kids. But the statues of Ty Cobb and Al Kaline did it for me. Those are in the outfield.

Comerica park doesn't have the surrounding neighborhood that Camden, Wrigley and Fenway have but there are some cools spots to hang out in before or after the game. My two favorites are Cheli's Chili Bar and Hockeytown Cafe. Cheli's has a rooftop area and multiple floors. There are also a lot of attractions in the Greektown neighborhood including the casino. You can stay there and not have to worry about parking anywhere in downtown Detroit since it's free to guests.

To give you an idea what the view of the field is like from section 326, here is a Youtube video shot from there during a Tigers/New York Yankees game at Comerica Park.
